摘要:本文利用了乙醇/磷酸氢二钾、乙醇/硫酸铵、聚乙二醇/硫酸铵双水相法分离提取了苦荞麦中的黄酮类化合物,通过比较黄酮类化合物的得率,选择双水相体系进行优化研究。同时,讨论了黄酮类化合物在双水相体系中的分配行为;也考察了超声波条件对黄酮类化合物提取的影响,通过正交实验选择最佳的提取工艺。主要内容如下:
1.研究表明,苦荞麦中提取黄铜类化合物得率最高的是乙醇与硫酸铵双水相体系。该体系上相中黄酮类化合物的得率为0.8439%,占上、下两相中黄酮类化合物总得率的94.92%。
2.通过硫酸铵的质量分数,乙醇的质量分数、以及pH值的讨论,对乙醇与硫酸铵双水相体系分配行为进行了研究。当乙醇的质量分数为35%,硫酸铵的质量分数为30%,pH值为8时,乙醇与硫酸铵双水相体系的分配系数达到最高。39447
3. 采用L¬9(34)对超声波辅助双水相萃取法提取苦荞麦中黄酮类化合物的工艺条件进行正交试验,优化提取工艺。得到最佳提取条件为:料液比为1:12、超声功率为420w,超声时间为3min,温度为50℃。在此条件下黄酮类化合物的得率为0.9446%。

Extraction of flavonoid in Fagopyrum Gaertn by aqueous two-phase sytem
Abstract:Works including the extraction of flavonoid in Fagopyrum Gaertn by aqueous two-phase system in this paper. The best aqueous two-phase system among ethanol/ dipotassium hydrogen phosphate, ethanol / ammonium sulfate and polyethylene glycol / ammonium sulfate was selected by comparing the absorbance. At the same time, not only the distribution behavior of flavonoids in the optimal aqueous two-phase system, but also the effection of extracting flavonoids under ultrasonic irradiatio were discussed to obtain the optimum technological .The main contents are as follows:
1. Ethanol-ammonium sulfate two-phase was chosen as the best aqueous two-phase system by comparing the different from the yield of flavonoids. The yield of flavonoids in the system was 0.8439%, accounting for 94.92% of the total flavonoids in the upper and lower two phases.
2.The partitioning behavior of flavonoids in aqueous two-phase of ethanol and dipotassiums hydrogen phosphate has been studied, and the influences of the concentration of dispotassiums hydrogen phosphate, pH value, concentration of ethanol on the distribution coefficents of the flavonoids have been discussed. The distribution coefficient of ethanol and ammonium sulfate aqueous two-phase system was the highest, when the mass fraction of ethanol was 35%, the mass fraction of ammonium sulfate was 30%, and the pH value was 8.
3. The extraction process was optimized by using the ultrasonic assisted method. And the selected optimal microwave conditions was chosen by comparing the different from conditions of the flavonoids yield. The solid-liquid ratio was 1:12, the ultrasonic power was 420w, the ultrasonic time was 3min, the temperature was 50℃. Under these conditions, the yield of flavonoids was 0.9446%.
