摘要: 为了解萝卜S受体蛋白激酶的结构和功能,本论文利用生物信息学方法对萝卜S受体蛋白激酶氨基酸组成、理化性质、亚细胞定位、跨膜区和信号肽以及二级结构等方面进行预测,并利用同源建模方法模建了三级结构特征,结合拉氏散点图评价了模建结果,预测了该蛋白质的活性位点信息。结果表明,S受体蛋白激酶由853个氨基酸组成;在其N端7-29位氨基酸存在1个跨膜区,方向由内向外,446-465位氨基酸处是一个由外向内的跨膜区,该蛋白是一个定位于细胞质中的分泌型蛋白,含有多个不同的磷酸化位点;二级结构以扩展束(占25.09%)和无规则卷曲(占47.48%)为主,存在14个不稳定区,4个潜在球形蛋白区;三级结构由14段α-螺旋和3组β-折叠构成,其中1组平行β-折叠片层结构由5个β折叠单链组成,位于蛋白质内部,另两组β-折叠片层结构由几个较小的β折叠单链组成,靠近蛋白质外部,是一个α+β类蛋白;该蛋白具有7种不同模体,可能参与不同生化反应或执行不同的功能;该蛋白与甘蓝、油菜等同种S受体蛋白激酶相似性较高,进化距离较近。以上这些结果为进一步研究S受体蛋白激酶在参与自交不亲和通路中的作用提供了理论基础。10200

Homology modeling,evaluation and pathway analysis of S receptor kinase in Raphanus sativus L
Abstract: To understand the structure and function of radish S receptor protein kinases, its amino acid composition, terms of physicochemical properties, subcellular localization, signal peptide, transmembrane domain and secondary structure were predicted by bioinformatics ways, then its tertiary structure was obtained by homology modeling, and the modeled structure qulity was estimated using Laplace scattergram; the protein active site of information was predicted. The results showed, S protein kinase receptor consists of 853 amino acids; presence of a transmembrane region at its N- terminal amino acids 7-29, the direction from the inside outward a,446 -465 amino acid transmembrane region is an outside, the protein is localized in the cytoplasm of a secreted protein, comprising a plurality of different phosphorylation sites; two structures to expand the beam ( 25.09 %) and random coil ( representing 47.48% ) there are mainly 14 unstable region, four potential globular protein region; tertiary structure of the α- helical segments 14 and three groups β- folded configuration, wherein a set of parallel β- pleated sheet structure of the folded single-chain β 5 , is located inside the protein the other two β- pleated sheet structure consists of several smaller single β chains folded , near the external protein is an α + β -like protein ; the protein has seven different mold , or may be involved in biochemical reactions performed in different different functions; the protein with cabbage , rape seed S is equivalent to the receptor protein kinases high similarity , evolutionary distance closer . These results further theoretical basis.
