摘 要: 在该研究中,里氏木霉发酵的纤维素酶被来降解预处理稻草,并以预处理稻草水解液作为解脂耶罗维亚酵母菌Yarrowia lipolytica SWJ-1b发酵产柠檬酸的原料。我们对Y. lipolytica SWJ-1b以预处理稻草水解液为碳源进行分批和补料发酵生产柠檬酸进行了研究。在分批培养中,以预处理稻草水解液为原料发酵柠檬酸产量为23.4 g/l,培养时间为120 h。而在以预处理草水解液为原料进行三轮的补料培养后,柠檬酸产量增加至42.4 g/l,培养时间延长至240h。在分批和补料培养中的细胞生长情况类似。发酵结束时,分批和补料培养的培养基中分别只有0.3 g/l和1.6 g/l的还原糖残余,这表明所添加的大部分碳源都被用于转化为柠檬酸和细胞量。54481

Abstract:In this study, crude cellulase that fermented from Trichoderma reesei was used to degrade the pretreated straw powder into pretreated straw cellulose hydrolysate.Batch and fed-batch production of citric acid from straw cellulose hydrolysate has been investigated using Yarrowia lipolytica SWJ-1b. In the batch cultivation, 23.4 g/l of CA was yielded from hydrolysate of straw cellulose, and the cultivation time was 120 h. while in the three-cycle fed-batch cultivation with straw cellulose hydrolysate, CA production was increased to 42.4 g/l and the cultivation time was extended to 240 h. The cell growth of SWJ-1b in fed-batch cultivation was similar to that during the batch cultivation. At the end of the batch cultivation and fed-batch cultivation, 0.3 g/l and 1.6 g/l of reducing sugar was left in the medium, suggesting that most of the added carbon was converted into CA and cell mass.

1 引言
柠檬酸(Citric acid,CA)是一种重要的有机酸,被广泛地用于食品,化学品,饮料业和制药业等众多工业领域[1]。工业柠檬酸主要通过黑曲霉(Aspergillus niger)以粮食为原料发酵所得[1],但目前发现黑曲霉能够引起肺曲霉肿等疾病,这成为柠檬酸发酵行业的重大安全隐患。基于此,具有高安全性的高产柠檬酸的解脂耶罗维亚酵母菌(Yarrowia lipolytica)引起人们的关注,Y. lipolytica主要分离自奶类肉类等食物,因而被公认为最安全的酵母菌。此外,该菌与A. niger相比还具有许多其他优点,如该菌能够耐受较高的重金属及污染环境,并且对低溶氧环境有较强的耐受性,因而可以利用许多工业粗原料发酵产柠檬酸,另外,其单细胞结构使得基于分子生物学手段的遗传改造更为简易,这些优点赋予了Y. lipolytica柠檬酸发酵领域的巨大潜力[2]。基于Y. lipolytica原料广泛的特点,降低发酵成本、提高发酵产量及优化发酵条件成为研究该菌发酵柠檬酸主要策略[2]。目前Y. lipolytica利用廉价原料发酵柠檬酸的报道铺天盖地,大量研究结果表明,在限制氮源的条件下,Y. lipolytica能够利用各种油类如植物油、椰枣糖浆、甘油、粗甘油、甘油副产物、乙醇、动物和植物脂肪发酵产柠檬酸[3],很多水果残渣如葡萄渣、菠萝废弃物[4]、猕猴桃果皮、果渣及蔬菜副产品如胡萝卜汁和芹菜渣[5]也被用于该菌柠檬酸的发酵生产,此外一些工厂废料如烷烃[6]、橄榄厂废水[7]等都被可用作Y. lipolytica发酵产柠檬酸的原料,这使得Y. lipolytica在降低柠檬酸发酵成本方面颇具优势。源'自:751`!论~文'网www.751com.cn
