摘要神经胶质瘤是最常见的原发性中枢神经系统肿瘤,严重威胁着人类的生命。因此为了能在临床上更好的了解脑瘤的生物学特性,进行筛选新药和检测药物的副作用,众多学者研究构建了多种动物肿瘤模型。其中根据肿瘤的来源可以将肿瘤模型分为诱发性脑胶质瘤模型、移植脑胶质瘤模型和转基因型脑胶质瘤模型。本实验构建的动物模型为移植脑胶质瘤模型。47372
目的 探讨SD大鼠C6胶质瘤细胞动物模型的建立。方法 借助动物活体成像仪的记录,采用脑立体定位仪微注射方法将体外培养大鼠胶质瘤细胞C6-Luc(悬浮于含有10%胎牛血清、1%双抗的DMEM培养液中),细胞数分别为1x106和2x106接种于SD大鼠左侧脑室,接种后观察肿瘤的生长状态。 结果 1x106细胞数和2x106接种于左侧脑室约10天后5只大鼠出现肿瘤,五只中3只最后成瘤存活,肿瘤在脑部形成明显肿瘤1周后发生转移,2周后肿瘤消失。 结论 本实验的鼠源胶质瘤细胞C6-Luc用于动物模型的建立成瘤率高,成瘤时间较适宜但易发生转移且一段时间后会消失,因此不适合作为筛选药物的脑肿瘤模型。
Abstract Glioma is the most common primary tumors of the central nervous system which has a great threat to human beings. In order to understand biological characteristics of brain tumor at the same time screening for new medicine and testing the detection of the side effects of medicinal properties, several of researchers establish different animal models of brain tumor. It pide into three kinds: induced brain glioma model, transplantation of glioma model, genetically modified glioma model. Our research wants to establish a transplantation of glioma model.
Purpose Make a discussion of establishment of C6 glioma model in SD rats. Methed With the help of animal in vivo imaging instrument recording, graft C6-Luc glioma cells(The cells’ number is 1x106 or 2x106, Suspended in the mixture including 10% FBS and DMEM nutrient solution with 1% penicillin and streptomycin) into rats’ left ventricle by micro injection methods using stereotaxic apparatus. Then observe and state the growth of tumor. Result 10 days after transplantation, the tumor grow in the ventricle. 3 rats survive at last and the tumor metastasize 1 week later . Finally the tumor disappear. Conclusion It has a high tumor formation rate using C6-Luc glioma cells and the tumor get to a suitable size in a suitable time although the tumor is easy to metastasis and finally disappear. So, a SD rat modle of C6 glioma cells is not a successful model.
