    摘要:TaRboh基因是植物体内调控抗胁迫的一类基因,存在于大多数体内,例如小麦,玉米等,这些植物在干旱,重金属,盐泽等非生物条件的胁迫下,便会引起该基因发挥作用,使植物产生一定抗性,因此研究该基因的功能有利于提高植物的抗逆性。本实验以模式植物拟南芥为实验材料,分别是TaRboh基因导入拟南芥体内的超表达拟南芥、敲除TaRboh基因的突变体拟南芥、还有野生型拟南芥三种类型。实验采用60umol/L浓度的镉盐处理拟南芥,通过观察处理前后以及不同类型拟南芥在受胁迫的情况下,其萌发、生长、形态指标的情况,通过测定拟南芥的各种生理指标含量,探究TaRboh基因对拟南芥受胁迫的作用,生理指标包括超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、脯氨酸、丙二醛(MDA)、过氧化物酶(POD)等。经过试验发现,超表达的拟南芥的生长发育情况最好,而且其酶活性也最高;野生型的生长发育相对突变体的较好,其酶活性也相对较高。因此通过本实验得出:TaRboh基因对模式之物拟南芥的受胁迫情况确实产生一定的作用,提高了拟南芥的抗逆性。42956

    Response of TaRboh gene to cadmium stress in Arabidopsis thaliana

    Abstract:TaRboh gene is a kind of gene that regulates the resistance to stress in plants, which is found in most of the body, such as wheat, corn, etc.These plants in drought, heavy metals, salt marsh and other abiotic conditions, will cause the gene to play a role, so that the plant has a certain resistance, so the study of the function of the gene is conducive to improve the resistance of plants.In this study, the model plant Arabidopsis thaliana as the experimental material, respectively, is the TaRboh gene into Arabidopsis thaliana in vivo overexpression of Arabidopsis thaliana, knocking out the TaRboh gene mutant Arabidopsis thaliana, there are three types of wild Arabidopsis thaliana.The cadmium salt 60umol/L concentration treatment of Arabidopsis thaliana, by observing the treatment before and after different types of Arabidopsis under stress, the germination, growth, morphological index, through the determination of the Arabidopsis various physiological indicators of content, explore gene TaRboh of pseudo Arabis stress,physiological parameters include superoxide dismutase (SOD), catalase (CAT), proline, malondialdehyde (MDA), peroxidase (POD), and so on.It was found that the growth and development of Arabidopsis thaliana was the best, and its ender the stress condition, which can improve the resistance of Arabinzyme activity was highest, and the wild type of growth and development was relatively good, and its enzyme activity was relatively high.Therefore, through this experiment, we can conclude that the TaRboh gene has a certain effect on the Arabidopsis thaliana udopsis thaliana.
