    摘 要:植物疾病抗性蛋白在植物防御疾病方面发挥着重要作用,对植物抗性蛋白的研究一直是各国科学家关注的重点。本文通过对油菜疾病抗性蛋白序列特性和进化展开分析,以期为进一步研究植物与病原菌的相互作用机制奠定一定的理论基础。本文从NCBI蛋白数据库中检索并下载了油菜疾病抗性蛋白序列,并采用生物信息学方法,对这些蛋白序列进行了多序列比对,同时对代表序列进行了理化性质、磷酸化、跨膜区、信号肽、亚细胞定位、保守域、二级结构、三级结构建模和进化等方面进行了预测和分析。结果表明,油菜疾病抗性蛋白序列包含20种基本氨基酸,与其他近缘物种的氨基酸序列有较高的相似性,该蛋白序列具有多个不同的磷酸作用位点,在N端不存在跨膜区和信号肽序列,属于非跨膜蛋白,亚细胞定位于细胞质中,该蛋白序列具TIR_2、AAA_ATPase 和PLNO3210保守域结构,存在多个较小的不稳定区,总体形成球型蛋白。此外,该蛋白的二级结构主要由α螺旋和无规则卷曲构成,是一个典型的α/β类蛋白。对该蛋白序列进行同源建模,Ramachandran 图检测表明此模型结构符合立体化学规则。32128

    The Characteristics And Evolution Analysis of Brassica Rape Disease Resistance Protein
    Abstract: Plant disease resistance protein plays an important role in plant defense against disease, which is a research hotspot in biology in recent years. In order to set a theoretical foundation for the further mechanism study in the interaction between  plant and pathogen, characteristics and evolution analysis for rape disease resistance protein sequence was conducted in the paper. The rape disease resistance protein sequence were download from the NCBI protein database using bioinformatics methods, such as multiple sequence alignment, the physical and chemical properties, phosphorylation prediction, transmembrane prediction, signal peptide prediction, subcellular localization analysi, conserved domain prediction, secondary structure analysis, tertiary structure modeling and evolution information were analyzed. The results showed that the disease resistance protein containing 20 basic amino acids, have high similarity in amino acid sequence with other homology species, the protein has multiple different phosphorylation sites, but with no transmembrane domain and signal peptide sequence at the N end, which belongs to non -transmembrane protein, the protein was located in the cytoplasm, they have TIR_2、AAA_ATPase and PLNO3210 conserved domain and there are many smaller unstable region, it take shape spherical protein on the whole. In addition, the secondary structure of the protein is mainly composed of alpha helix and random coil which is a typical alpha / beta proteins. Ramachandran detection showed that this model accords with the rules of stereochemistry qulity.
