    摘要:本文以黑苦荞茶为原料,主要研究了通过乙醇提取法、超声波提取法与改造型索氏提取器法提取黑苦荞茶中的黄酮类物质,通过单因素试验和响应面试验确定了前两种提取方法的最佳工艺条件,并对三种提取方法进行了比较,利用大孔吸附树脂D101纯化黑苦荞茶黄酮类物质的粗提液,并以芦丁标准品与文C为对照,研究了黑苦荞茶黄酮类物质纯品与粗品清除DPPH•的能力。
    主要试验结果如下:
        1、乙醇溶液浸提黑苦荞茶黄酮类物质的最佳工艺条件为:固液比为1:30g/ml,提取温度为80℃,乙醇浓度为60%,提取5min,提取1次,其提取率为1.799%。
        2、超声波辅助提取黑苦荞茶黄酮类物质的最佳工艺条件为:固液比为1:30g/ml,超声功率为600w,乙醇浓度为30%,提取5min,提取1次,其提取率为2.029%。
        3、通过对乙醇提取法、超声波提取法与改造型索氏提取器法三者的最佳提取工艺条件下的提取效果的比较,发现提取效果最优的为改造型索氏提取器法(提取率为2.168%),超声波提取法(提取率为2.029%)次之,最差的是乙醇提取法(提取率为1.799%)。22198
        4、通过对DPPH•清除效果的研究,发现黑苦荞茶黄酮类物质对DPPH•具有较好的清除效果,且清除率随其浓度的增加而增强。在清除率最高时,黑苦荞茶黄酮类物质纯品与粗品的清除率分别为91.93%与87.95%,而对照组的芦丁标准品与文C的清除率分别为94.96%与50.40%。

    Study on Extraction Conditions and Antioxidant Activity of Flavoniods from Tartary Buckwheat Tea
    Abstract:The study used tartary buckwheat tea as material, mostly researched the extraction methods of ethanol, ultrasonic and the reformed Soxhlet extractor. The optimal parameters of the first two extraction methods was identified by single factor and RSA experiment, meantime, these three methods were compared. On the other hand, one type of macroporous resins: D101 was used in filtering out crude goods of flavoniods from tartary buckwheat tea. Finally, the antioxidant activity of pure and crude goods of flavoniods from tartary buckwheat tea was studied in DPPH• dilute comparing with rutin standard solution and vitamin C.
        The main results showed:
        1.The optimal parameters of ethanol extraction from tartary buckwheat tea were as following: material/liquid ratio is 1:30g/ml, temperature is 80℃, ethanol concentration is 60%, extracting 5 min and one times,its extraction rate is 1.799%.
        2.The optimal parameters of ultrasonic extraction from tartary buckwheat tea were as following: material/liquid ratio is 1:30g/ml, ultrasonic power is 600w, ethanol concentration is 30%, extracting 5 min and one times, its extraction rate is 2.029%.
        3.By contrasting these three methods: ethanol extraction, ultrasonic extraction and the reformed Soxhlet extractor’s extraction optimum extraction technology, we found the optimal extraction method is the reformed Soxhlet extractor(extraction rate is 2.168%), ultrasonic extraction method(extraction rate is 2.029%) followed, and ethanol extraction(extraction rate is 1.799%) was the worst.
        4.By studying on flavonoids’ DPPH• scavenging, we discovered that flavoniods from tartary buckwheat tea does have good cleaning effect of DPPH•, and its DPPH • scavenging increasing with its concentration increased. Most of all, in the highest scavenging, the DPPH•(0.5×l0-3mol/l) scavenging of pure and crude goods of flavoniods from tartary buckwheat tea were 91.93% and 87.95%, while the control group with vitamin C and rutin standard’s DPPH•(0.5×l0-3mol/l) scavenging were 94.96% and 50.40% .
